新疆边境地区蜱携带梨形虫种类的分子鉴定

摘  要:
为了调查新疆边境地区梨形虫在当地蜱中的携带情况,对采集于2019年4月份和5月份的新疆边境地区共25个县(市)的蜱进行形态学鉴定,提取DNA,利用梨形虫套式引物进行扩增并测序,并采用IQ-Tree软件的ML法构建不同地理株的进化关系。结果显示,采集到的1 516只蜱包括4个属9个种,分别为森林革蜱(Dermacentor silvarum)、草原革蜱(D.nuttalli)、胫距革蜱(D.pavlovskyi)、边缘革蜱(D.marginatus)、亚洲璃眼蜱(Hyalomma asiaticum)、麻点璃眼蜱(H.rufipes)、小亚璃眼蜱(H.anatolicum)、血红扇头蜱(Rhipicephalus sanguineus)和刻点血蜱(Haemaphysalis punctate);对上述蜱种携带梨形虫种类的分子检测结果显示,共检出3种泰勒虫和6种巴贝斯虫,分别为绵羊泰勒虫(Theileria ovis)、马泰勒虫(T.equi)、泰勒虫未定种(Theileria sp.)、驽巴贝斯虫(Babesia caballi)、粗糙巴贝斯虫(B.crassa)、隐藏巴贝斯虫(B.occultans)、犬巴贝斯虫(B.canis canis)、莫氏巴贝斯虫(B.motasi)及巴贝斯虫未定种(Babesia sp.)。结果表明,蜱中梨形虫的平均携带率为27.77%,且新疆边境不同地区的蜱梨形虫的携带率不同。研究结果为梨形虫病的进一步防控以及相关措施的制定提供了依据。

摘  要:
The purpose of this study was to investigate the piroplasma in ticks of Xinjiang border areas, we collected and identified ticks from 25 counties(cities) of Xinjiang border areas from April to May in 2019.The gDNA of ticks were extracted, then the extracted genome was amplified and sequenced by using primers of piroplasma in PCR.The ML method of IQ-Tree was used to construct the molecular phylogenetic relationship in various regions.The result showed that we collected 1516 ticks, including nine species of four genera, which were identified as: Dermacentor silvarum,D.nuttalli,D.marginatus,D.pavlovskyi,Hyalomma asiaticum,H.rufipes,H.anatolicum,Rhipicephalus sanguineus and Haemaphysalis punctate;the PCR result indicated three Theileria species and six Babesia species were detected in these ticks, including Theileria ovis,T.equi,Theileria sp.,Babesia caballi,B.crassa,B.occultans,B.canis canis,B.motasi and Babesia sp..The positive rate of piroplasma in ticks was 27.77%,and the positive rate of piroplasma was different in different border areas of Xinjiang.The result provides a basis for further prevention and control of piroplasm.
