沟眶象纤维素酶性质的研究

摘  要:
对沟眶象成虫肠道内纤维素酶的组成、性质和水解动力学特征等进行研究.结果表明,沟眶象成虫肠道内有完整的纤维素酶系,其中以内切β-l,4-葡聚糖酶(Cx酶)活性最强,外切β-l,4-葡聚糖酶(C1酶)次之,β-l,4-葡萄糖苷酶的活性最弱;Cx酶、C1酶的最适作用温区分别为30~55℃和40~60℃,最适作用温度分别为45℃和55℃;最适pH值分别为5.6和5.0;Cx酶具有最强的热稳定性,C1酶热稳定性较弱;动力学参数比较显示,Cx酶的最大反应速度(vmax)和米氏常数(Km)均最大,而β-l,4-葡萄糖苷酶的Km和vmax均最小.

摘  要:
The character and composing of cellulase in Eucryptorrhynchus chinenis(Olivier) are studied.The results showed that the Eucryptorrhynchus chinenis(Olivier) had integrated cellulolytic enzyme complex :exo-β-1,4-glucanase(C1-ase),endo-β-1,4-glucanase(Cx-ase) and β-1,4-glucosidase in their gut,among which the activity of Cx-ase was the strongest and β-1,4-glucosidase was the weakest.For Cx-ase and C1-ase,the optimum temperature was 45 ℃ and 55 ℃,and the optimum temperature range was between 30~55 ℃ and 40~60 ℃ respectively,while their optimum pH at 5.6 and 5.0.Cx-ase had the stronger stability to heat,but C1-ase was very weak.After comparison of the kinetic parameters among the cellulases of Eucryptorrhynchus chinenis(Olivier),it was concluded that Cx-ase had greater vmax and Km.
