氯化钙对重组运动发酵单胞菌生产乙醇的影响

摘  要:
为了提高乙醇产量,采用基因克隆的方法,将T7表达系统与fadK基因组整合到运动发酵单胞菌(Zymomonas mobilis)基因组中,获得能高产乙醇的重组工程菌株。在糖浓度为24%的玉米水解液发酵培养基中,重组工程菌株Z.M.F-4的产乙醇量比对照菌株提高了7%。在不同氯化钙浓度的发酵培养基中,重组菌Z.M.F-4发酵生成的乙醇产量最高的氯化钙浓度为16mmol/L,其乙醇产量为(101.50±3.59)g/L。在此钙离子浓度中添加1‰玉米油发酵,其乙醇产量为(101.10±3.73)g/L,与不添加玉米油时的乙醇产量几乎一致。

摘  要:
In order to enhance ethanol production,the T7-expression fadK genomes were introduced into Z. mobilis to get recombinant bacteria with high ability of ethanol production by gene cloning methods. However,compared with Z.mobilis ATCC 3182,there was 7% increasing of ethanol production for the recombinant bacteria named as Z.M.F-4 with the corn hydrolyaztes contained 24% sugar as fermentation medium. The 16 mmol/L supplement of calcium chloride could get the optimal ethanol yield which was (101.5±3.59)g/L from a range concentration of calcium chloride. Add to 1 ‰ corn oil in this fermentation medium with 16 mmol/L calcium ion,the ethanol yield was (101.1±3.73) g/L,which was almost the same as non-corn oil medium.
