摘 要:
试验设置水分散粒剂(H14)、商业可湿性粉剂(SY)和空白对照(CK)3组处理,在棉花花铃期调查黄萎病发生状况,并通过高通量测序分析不同处理中细菌群落多样性。结果显示同CK处理相比,H14和SY处理中棉花黄萎病发病率分别显著下降42.35%和39.25%,病情指数分别显著下降52.41%和48.78%;大丽轮枝菌(Verticillium dahliae Kleb.)相对丰度分别显著下降45.73%和24.47%。3组处理样品测序后共获得517 453个优质序列,基于97%的序列相似度进行聚类分析,获得可操作分类单元(OTUs)24 353个,经鉴定属于细菌的54个门、244个科和571个属。其中变形菌门(Proteobacteria)、芽单胞菌门(Gemmatimonadetes)、放线菌门(Actinobacteria)、酸杆菌门(Acidobacteria)和绿弯菌门(Chloroflexi)为各处理样品的优势菌群。H14、SY处理中Chao1和ACE指数相较于CK处理分别增加16.27%和14.71%、13.84%和12.90%;Shannon和Simpson指数相较于CK处理分别增加2.65%和1.02%、1.38%和0.00%。全部样品丰度前35个属通过聚类分为A、B、C、D 4个类群,其中C和D两个类群是H14处理中优势类群,此类群中的芽孢杆菌属(Bacillus)、芽生球菌属(Blastococcus)和类诺卡氏菌属(Nocardioides)多为生防菌;斯科曼氏球菌属(Skermanella)、亚硝化螺菌属(Nitrosospira)多为氮循环功能菌;酸杆菌属(Acidibacter)和土壤红杆菌属(Solirubrobacter)多为产铁载体促生菌。施用水分散粒剂H14可以提高棉花根围土壤细菌群落物种丰富度和多样性,增加防治土传病害以及氮铁等营养元素转化的相关菌属丰度,有利于改善土壤生态环境,抑制大丽轮枝菌繁殖,促进棉花健康生长。

摘 要:
The soil bacterial community is important for healthy cotton growth three treatments water dispersible granules H14(Bacillus mojavensis),commercial wettable powder(SY) and control were set to survey the progression of Verticillium wilt of cotton,furthermore,how H14 improved the performance against Verticillium wilt of cotton by affecting soil bacterial community diversity was studied in the paper.This is potential to provide theoretical guidance for uses of microbial inoculum in field.After separate root irrigation with water dispersible granules H14,commercial wettable powder and water,Verticillium wilt occurrence and collection of soil samples from cotton rhizosphere in the flowering and boll stage respectively were quantified,then,the diversity of soil bacterial community under different treatments by high throughput sequencing was analyzed.The disease incidence of cotton Verticillium wilt in H14 and SY treatments significantly decreased by 42.35% and 39.25% respectively compared with the control,while disease index significantly decreased by 52.41% and 48.78% respectively.The relative abundance of Verticillium dahliae Kleb.in H14 and treated soil by SY significantly decreased by 45.73% and 24.47% respectively compared with the control.A total of 517 453 high quality sequences were obtained from all samples.And a total of 24 353 operational taxonomic units(OTUs) of all samples were obtained by cluster analysis according to 97% sequence similarity,which were classified into 54 phyla,244 family and 571 genus.The dominant bacteriophyta were Proteobacteria,Gemmatimonadetes,Actinobacteria,Acidobacteria and Chloroflexi in all samples.The Chao1 and ACE index with H14 and SY treatments increased by 16.27% and 14.71%,13.84% and 12.90% respectively compared with the control,while the Shannon and Simpson index increased by 2.65% and 1.02%,1.38% and 0.00% respectively.The top 35 genera in all samples were divided into four groups,i.e.,A,B,C,and D.The dominant bacteriogenus were C and D groups in the treatment of water dispersible granules H14,which were composed of bicontrol bactreia( Bacillus,Blastococcus,Nocardioides),nitrogen cycling functional bacteria(Skermanella,Nitrosospira) and siderophore-producing bacteria(Acidibacter,Solirubrobacter).The results predicted that water dispersible granules H14 could improve the diversity of cotton soil bacterial community and inhibit the reproduction of Verticillium dahliae Kleb. finally,it was benefit to healthy cotton growth.
