摘  要:
酶的定向进化是用于改良酶特性的一种策略,主要包括构建酶突变文库和高通量筛选。目前,突变文库的构建手段主要有随机突变、半理性设计和DNA改组;高通量筛选方法主要分为体内筛选和体外筛选。该文介绍易错PCR、半理性设计、DNA改组、表面展示、核糖体展示和差示荧光扫描等相关技术及其应用情况。

摘  要:
Direct evolution of enzyme,a strategy to improve the characteristic of enzyme,mainly included constructing enzyme mutagenesis library and high-throughput screening. At present,the main methods of constructing enzyme mutagenesis library were random mutation,semi-rational design and DNA shuffling; high-throughput screening can be divided into in vivo screening and in vitro screening. In this study,the related techniques and their application situation,including error-prone PCR,semi-rational design,DNA shuffling,surface display,ribosome display and differential scanning fluorimetry,were introduced.
