内生固氮菌HAUMIO对水稻的侵染定殖规律及促生效应

摘  要:
试验以从表面灭菌的稗草中分离到的一株内生固氮菌HAUM10为对象,研究发现该菌具有较强的产固氮酶和吲哚乙酸能力.乙炔还原法测定其所产固氮酶的活性为0.824μmol C2H4/(mL·h),液相色谱法测定其产吲哚乙酸量为15.44μg/mL.将HAUM10用疥基因标记后接种水稻,观察其在水稻中的侵染定殖规律,发现在接种后第7天,加标记的HAUM10主要定殖于根内通气组织和韧皮部细胞,少量定殖在叶鞘中.盆栽试验表明,HAUM10促进水稻叶绿素合成,有利于光合作用,在水稻生长后期促进氮、磷元素由叶向穗转移.

摘  要:
An endophytic diazotroph HAUM10 was originally isolated from surface-sterilized Echinochloa crusgalli and it pre- sented appreciable levels of nitrogenase activity and IAA production capability. The nitrogenase aetivity(ARA) was about 0.824μmol C2H4/(mL·h). The liquid chromatography method was performed to determine the 1AA production of HAUM10 which was detected as up to 15.44 μg/mL. Infection and colonization of the rice seedlings by gfp-tagged HAUM10 were examined. HAUM10-gfp was mainly colonized within the root aerenchyma and phloem at the 7th day after inoculation; While a few was observed in the leaf sheath. Pot experiment showed that the strain of HAUM10 increased the chlorophyll content to promote the photosynthesis of rice and transported the nitrogen and phosphorus from leaf to spike in the late growth of rice.
