猪红细胞类补体受体Ⅰ型膜结合蛋白的筛选

摘  要:
本研究旨在鉴定猪红细胞类补体受体Ⅰ型(complement receptor 1-like,CR1-like)的膜结合蛋白,以期为探讨CR1-like在膜上分布的分子基础提供理论数据。采用免疫磁珠沉淀的方法纯化猪红细胞CR1-like,利用还原性SDS-PAGE电泳及Western-blot试验,得出猪红细胞CR1-like的分子大小;利用质谱技术对纯化的CR1-like免疫沉淀复合物进行鉴定,并对质谱数据进行相关分析。CR1-like免疫沉淀复合物的还原性SDS-PAGE和Western-blot的鉴定结果显示,CR1-like分子大小约为50 ku;CR1-like免疫沉淀复合物的质谱鉴定筛选出117个候选蛋白,根据肽段数、特异肽段数、序列覆盖率、分值等及对候选蛋白的生物信息学分析筛选出FERMdomain-containing protein作为候选蛋白进行下一步试验。上述结果表明,本研究成功建立了免疫磁珠沉淀CR1-like方法,并筛选出CR1-like的膜结合候选蛋白。

摘  要:
To provide theoretical data for exploring the molecular basis of CR1-like distribution on erythrocyte membranes,we identified CR1-like membrane-binding proteins on porcine erythrocytes.Porcine erythrocytes were puried by immunomagnetic beads precipitation for CR1-like and the molecular weight was determined using SDS-PAGE and Western-blot in reducing condition.Furthermore,CR1-like complex was identified by mass spectrometry and the datas obtained were analyzed.The results of SDS-PAGE and Western-blot showed that the molecular weight of CR1-like is about 50 ku.117 candidate proteins were screened out by mass spectrometry and FERM domain-containing protein was the candidate protein obtained which was screened out for subsequent research according to the number of peptides,the number of special peptides,the sequence coverage,score and bioinformatics analysis.In conclusion,the method of immunomagnetic beads precipitation for CR1-like was established and the candidate membrane-binding protein(FERM domain-containing protein) for CR1-like was screened out successfully.
