抗猪红细胞膜抗原非凝集型单克隆抗体的研制及鉴定

摘  要：
以猪红细胞膜抗原免疫BALB/c小鼠,应用杂交瘤技术将免疫小鼠的脾细胞与Sp2/0骨髓瘤细胞融合,通过微孔板凝集试验及玻片凝集试验筛选可稳定分泌非凝集型mAb的杂交瘤细胞株,并用有限稀释法进行克隆。制备出一株可稳定分泌非凝集型抗体的杂交瘤细胞株1C2,其亚型为IgG1类,培养上清和腹水效价分别为1∶1024和1∶108,没有种属交叉血凝反应,为构建双特异性抗体奠定了基础。

摘  要：
BALB/c mice were immunized with the membrane antigens of swine erythrocytes.The splenocytes of the immunized mice were fused with Sp2/0 myeloma cells by hybridoma technique.The antibodies to common antigen of swine erythrocytes were screened by micro-quantity plate agglutination test method and then the agglutinating antibodies were weeded out by slide hem-agglutination assay.The hybridoma cells secreting non-agglutinating antibodies were cloned by end dilution method.One hybridoma cell 1C2 was obtained,which secreted non-agglutinating antibody.The mAb 1C2 belonged to IgG1,and without specie scross-agglutination reaction.The titers of cultures suppernatant and ascites of 1C2 were 1∶1024 and 1∶108 respectively.The mAb 1C2 has high titer,affinity and specificity,which could be used for the preparation of bi-specific antibodies(BsAb).
