水牛CART基因的克隆测序及蛋白质结构与功能预测

摘  要:
为探讨水牛可卡因-苯丙胺调节转录肽(cocaine-and amphetamine-regulated transcript, CART)基因功能,以水牛卵巢组织RNA转录的cDNA为模板,参考GenBank预测的水牛CART基因组序列设计引物,采用PCR和在线生信分析软件对CART基因进行克隆和蛋白质结构预测分析。结果显示:水牛CART基因CDS区全序列长为351 bp,编码116个氨基酸;对所得核酸序列和蛋白质结构分析预测显示,水牛CART基因与美洲野牛、野牦牛、普通牛、羊、双峰驼、单峰驼、野猪、马、非洲野驴、人类及猕猴相似程度分别为100%、98%、100%、98%、91%、91%、78%、85%、85%、81%、90%;Mega分析显示在多胃反刍动物间有较高同源性;蛋白质结构预测显示,CART蛋白含有N端信号肽,是分泌蛋白,功能与神经、辅酶和离子结合相关。

摘  要:
In this study, the cocaine-and amphetamine-regulated transcript(CART) gene of buffalos was cloned and its protein structure was predicted. The cDNA transcribed by RNA was extracted from the ovaries of buffalos as templates, primers were designed according to the genomic sequence of CART predicted by GenBank so as to amplify and sequence the CDS region of the CART gene of theanimals. The results showed that the total length of the CDS region of the buffalo CART gene was 351 bp, encoding 116 amino acids. The analysis of nucleic acid sequence and protein structure indicated that the similarities between the buffalo CART gene and that of American bison, wild yak, common cattle, sheep, bactrian camel and single hump camel, wild boar, horse and African wild donkey, human and macaque were 100%, 98%, 100%, 98%, 91%, 91%, 78%, 85%, 85%, 81% and 90%, respectively. The prediction of protein structure indicated that the CART protein contained N-terminal signal peptides confirming it to be a secretory protein. The present study laid a foundation for further research on the function of the CART gene in buffalos.
