斑嘴鸭线粒体全基因组序列测定及分析

摘  要:
根据斑嘴鸭(Anas poecilorhyncha)同属近缘物种绿头鸭(Anas platyrhynchos)线粒体基因组(mt DNA)序列设计引物,采用直接测序技术对斑嘴鸭mt DNA全序列进行综合分析。结果显示,斑嘴鸭mt DNA序列全长16 606 bp,包含22个tRNA、2个rRNA基因、13种蛋白质编码基因和1个D-loop区。碱基组成T为22.2%,C为32.8%,A为29.2%,G为15.8%,无明显的AT偏好性,22种tRNA都为典型的三叶草结构。参照棕头鸦(Larus brunnicephalus)、黑尾地鸦(Podoces hendersoni)的12S rRNA,对斑嘴鸭12S rRNA的二级结构进行预测,结果其含有4个结构域,37个茎环。对D-loop控制区序列分析发现含有goose hairpin,E-box,F-box,D-box,Bird similarity-box及CSB1-box。并以原鸡作为外群,采用N-J算法和ML算法,基于mt DNA全序列及D-loop区分别构建系统进化树,发现3个家鸭品种与绿头鸭亲源关系较近。

摘  要:
According to Spotbill( Anas poecilorhyncha) closely related species Mallard( Anas platyrhynchos),primers were designed,using direct sequencing techniques. Spotbill mitochondrial genome sequence were analyzed comprehensively,Anas poecilorhyncha whole mitochondrial genome sequence is 16 606 bp,contains 22 tRNA,2 rRNA genes,13 protein-coding genes and a D-loop region. Base composition T was22. 2%,C was 32. 8%,A was 29. 2% and G was 15. 8%,which was no significant AT preference. 22 kinds of tRNA were typically cloverleaf structures. Referring to 12 S rRNA of brown head crow( Larus brunnicephalus) and black-tailed ground crow( Podoces hendersoni),the secondary structure of the spot-billed ducks were predicted,finding 12 S rRNA contains four domains,37 a stem-loop. The D-loop control region was found with goose hairpin,E-box,F-box,D-box,Bird similarity-box and CSB1-box. And taking the original chicken as an outgroup,using N-J algorithm and ML algorithm,based on mitochondrial genome sequences and D-loop region phylogenetic tree was constructed and found three domestic ducks breed and Anas platyrhynchos had closer relationship.
