BMPR-IB基因作为多浪羊高繁殖力候选基因的研究

摘  要:
【目的】以骨形态发生蛋白受体(BMPR-IB)基因作为多浪羊的高繁殖力候选基因,研究该基因对多浪羊产羔性状的影响。【方法】采用PCR-SSCP和PCR-RFLP技术检测多浪羊BMPR-IB基因的单核苷酸多态性;用最小二乘法分析不同基因型与产羔数的相关性;并对基因型纯合个体进行测序。【结果】BMPR-IB基因在多浪羊中存在三种基因型即野生型++、突变杂合型B+和突变纯合型BB;基因型频率分别为0.741、0.254和0.005;B和+等位基因频率分别为0.132和0.868。BB基因型与++基因型相比在BMPR-IB基因编码区第746位碱基处发生了和Booroola绵羊相同的突变(A→G)。B+基因型的平均产羔数比++基因型多0.51只(P<0.01),差异极显著。【结论】BMPR-IB基因是影响多浪羊多胎性状的一个主效基因,可作为一个分子标记用于多浪羊的选育。

【Objective】Bone morphogenetic protein IB receptor(BMPR-IB)gene as a candidate gene for prolificacy in Duolang sheep was studied for its effect on the lambing character of Duolang sheep.【Method】Using PCR-SSCP and PCR-RFLP technologies, single nucleotide polymorphism of BMPR-IB gene in fecundity sheep breed Duolang sheep was studied.The relationship between genotypes and litter size was analysed by means of the Least-squares method.And the homozygous was sequenced.【Result】BMPR-IB gene had three genotypes in Duolang sheep: Wild-type ++,Heterozygous mutation B+ and Homozygous mutation BB;the genotype frequency was: 0.741,0.254 and 0.005 respectively,and the gene frequency of B allele and + allele were: 0.132 and 0.868.Compared with the++genotype,the sequencing results revealed a same mutation(A→G) at 746 bp of BMPR-IB gene in the BB genotype of Duolang sheep as in Booroola sheep.Duolang sheep ewes with genotype B+ had 0.51(P<0.01) more lambs than those with genotype ++ in average.The difference was extremely significant.【Conclusion】BMPR-IB gene was a major gene that affected the prolificacy in Duolang sheep,and could be used as a molecular marker to select Duolang sheep.
