摘  要:
分子定向进化作为近几年发展起来的一种蛋白质改造新策略,由编码基因的随机突变、重组和定向筛选构成。在不了解蛋白的三维结构信息和功能机制的情况下,利用这一技术调整目标蛋白的性质,如专一性、催化活性及亲和力等。近年来在易错PCR和DNA shuffling的基础上已经出现了很多新的分子定向进化技术。概述了易错PCR和DNA shuffling的原理及其发展与应用。

摘  要:
Directed evolution,based on the combination of random mutagenesis,recombination and high throughput screening,is a new strategy of improving protein properties.With this technology,the properties of the target proteins,such as specificity,catalytic activity and affinity can be tuned without the knowledge of 3-D structure information and function mechanisms.Recently,the methodology originating from error prone PCR and DNA shuffling has been greatly developed by many new techniques.In this paper,the principles of error prone PCR and DNA shuffling and their innovation,development and application were introduced.
