中国石竹叶片愈伤组织形成·分化及植株再生

摘  要:
[目的]为了研究中国石竹叶片愈伤组织的诱导及植株的再生。[方法]通过叶片愈伤组织诱导再分化和叶片直接诱导不定芽2种途径获得了石竹的再生植株。[结果]以植株中部带叶基的叶片愈伤组织诱导再分化效果最好。叶片愈伤组织在MS+6-BA 1.0mg/L+NAA 0.3mg/L培养基中不定芽分化率最高(50%~60%)。幼叶在MS+NAA 0.3mg/L+6-BA1.0mg/L培养基上培养47d可直接诱导出不定芽。与愈伤组织分化成芽相比,幼叶直接诱导不定芽芽数少,但生长快,苗较高。最佳增殖培养基是MS+6-BA 0.2mg/L+IAA 0.2mg/L,最佳生根培养基是1/2MS+IBA 1.0mg/L+NAA 0.1mg/L+0.2%活性碳。[结论]叶片作为外植体具有来源丰富、取材方便、操作简单的优点,探讨植物叶片的再生分化特性具有实际的意义。

摘  要:
Objective The research was conducted to investigate the induction of leaf callus and plant regeneration of Dianthus chinensis L.Method The regeneration plant of Dianthus chinensis L.was obtained by two ways of the leaf callus redifferentiation and the adventitious buds induced directly by lamina.Result The leaf callus from the middle plant with leaf base got best effect on inducing redifferentiation.The leaf callus on the medium of MS+6-BA1.0 mg/L +NAA0.3 mg/L obtained the highest differentiation rate of adventitious buds,being 50 %~60 %.The adventitious buds could be directly induced from young leaf when they were cultured on the medium of MS+NAA0.3 mg/L+BA1.0 mg/L for 47 days.Compared with differentiation bud from the callus,the adventitious buds induced directly from young leaf had few buds,but grew quickly and was higher in seedling height.The optimum proliferation medium was MS+6-BA0.2 mg/L+IAA 0.2 mg/L and the optimum rooting medium was 1/2MS+IBA1.0 mg/L+NAA 0.1 mg/L+ 0.2 % activated carbon.ConclusionThe leaf as explant had the advantages of rich resource,convenient sampling and simple operation.It had a practical significance to discuss the regeneration and differentiation characteristics of plant leaf.
