三七总皂苷粗提物S-3溶血性及体外免疫活性的研究

摘  要:
经大孔吸附树脂分离纯化,从三七中得到三七总皂苷(PNS) S-3组分,研究其溶血性及体外免疫活性。采用红细胞溶血率测定溶血性,以体外小鼠T、B淋巴细胞增殖反应及IL-2诱生检测S-3的体外免疫活性。结果表明:S-3浓度在0.25 mg·mL~(-1)以下时无溶血性,在125μg·mL~(-1)以下时未表现出对脾淋巴细胞活力有抑制作用。S-3浓度为10μg·mL~(-1)时对刀豆蛋白A诱导的体外小鼠T淋巴细胞增殖有极显著促进作用,为100μg·mL~(-1)时则极显著抑制T淋巴细胞增殖反应;而PNS浓度在1~100μg·mL~(-1)范围内均呈现出显著促进作用, S-3浓度为10、100μg·mL~(-1)时极显著抑制LPS诱导的小鼠B淋巴细胞增殖作用,在此浓度下PNS显示出对B淋巴细胞增殖有显著或极显著促进作用。S-3浓度为25μg·mL~(-1)时极显著促进小鼠T淋巴细胞IL-2分泌,与相同浓度的PNS相比, S-3无更显著的促进作用。这一研究提示S-3具有较好的安全性,且在体外具有一定的细胞免疫活性和诱生细胞因子的能力,为从PNS中开发出安全、高效的动物免疫增强剂奠定了基础。

摘  要:
The hemolytic characteristics and immune modulating activity of Panax notoginseng saponins(PNS) S-3 in vitro were investigated in this study. PNS was extracted with general method of normal butyl alcohol. S-3 was obtained by separating and purifying PNS with D-101 macroreticular resin. The hematolysis activity, the range of safe dose and the immune activity of S-3 in vitro were detected with hemolysis test, the content determination of IL-2 in T lymphocytes in mice, experiment of solenocytes toxicity and splenocyte proliferation respectively. The results showed that S-3 had no hemolytic characteristics at the concentration of less than 0.25 mg·mL~(-1), and no inhibitory effect on spleen lymphocyte activity at less than 125 μg·mL~(-1). S-3 increased significantly concanavalin A-stimulated splenocyte proliferation in vitro at the concentration of 10 μg·mL~(-1)and inhibited significantly it at 100 μg·mL~(-1), but PNS increased significantly it at the range of 1-100 μg·mL~(-1); S-3 inhibited significantly lipopolysaccharide-stimulated splenocyte proliferation in vitro at 10 and 100 μg·mL~(-1), but PNS increased significantly it at the same concentrations. S-3 increased significantly the content of IL-2 at 25 μg·mL~(-1) and had no more significant increase than PNS. In conclusion, S-3 was safe and reliable, and could be further studied and developed for an immunopotentiator in breeding of livestock.
