拮抗放线菌Sy11菌株感应的致病疫霉信号分子初步研究

摘  要:
为明确拮抗放线菌Sy11菌株在预防马铃薯晚疫病方面的潜力,以及Sy11菌株与致病疫霉间的感应现象,采用打孔法和离体组织培养等方法检测Sy11菌株对致病疫霉的抑制和离体生防效果,通过对峙培养法和纸板隔离法明确菌株间的感应现象以及由Sy11菌株感应的致病疫霉信号分子的溶解性和稳定性。结果表明,Sy11菌株活体和菌液对致病疫霉菌丝体生长的抑制率分别为89.7%和82.5%,菌液在马铃薯离体块茎和叶片上的相对保护率均高于85.0%;此外Sy11菌株只有在致病疫霉的诱导作用下才分泌抑菌物质,诱导后产生并分泌的抑菌物质的抑菌率达76.5%;Sy11菌株感应的致病疫霉信号分子易溶于甲醇,可耐受80℃高温。以上结果表明Sy11菌株在预防马铃薯晚疫病方面有较大潜力。本研究为今后进一步分离纯化致病疫霉信号分子,明确感应诱导作用机理提供参考依据。

摘  要:
In order to identify the potential of Sy11 strain in preventing potato late blight and the response between actinomycetes Sy11 strain and Phytophthora infestans, the punch method and detached tissue method were adopted to evaluate the inhibition of Sy11 strain on P. infestans and the prevention of late blight on potato tuber slices and leaves. The dual-culture method and cardboard isolation method were used to affirm the response between both strains and to clear the solubility and stability of signal molecule produced by P. infestans to Sy11 strain response. The results indicated that inhibitory rates of Sy11 strain living body and mycelium liquid to P. infestans mycelium growth were 89.7% and 82.5%, respectively. The relative protection ratios of Sy11 mycelium liquid on both in vitro tuber slices and detached leaves were higher than 85.0%. In addition, it was verified that under the induction of P. infestans only, Sy11 strain could secrete antiomycetes substances and its inhibitory rates could reach up to 76.5%. Signal molecule from P. infestans responded by Sy11 strain soluble in methanol and tolerant 80℃ temperature. These results not only indicate the prominent potential of Sy11 strain on prevention potato late blight, but also provide a reference for further separation and purification of P. infestans signal molecule as well as the clarification of mechanism of response and induction.
