RAPD技术在彩棉品种鉴定中的应用

摘  要:
以45份棉花材料(彩棉21份,白棉24份),利用改良的CTAB微量提取法从无菌幼苗中提取总DNA,进行RAPD扩增,以发现供试彩棉材料的特征谱带。研究表明,已筛选出RAPD随机引物S2032可使新彩棉4号品种引起特异性扩增,该随机引物扩增效果稳定、清晰,对45份材料进行RAPD电泳图谱分析,各材料间图谱差异明显,易于将新彩棉4号品种从45份材料中区分开。结果证明RAPD技术应用于彩色棉品种鉴定和纯度分析是可行的,并奠定利用RAPD技术对彩色棉品种进行鉴定和种子纯度分析的基础。

摘  要:
In this experiment, the total DNA of 45 cottons (21 colored cotton, 24 white cotton) are extracted by improved CTAB method, we are trying to find the typical pedigree printing of colored cotton by the random amplified polymorphic DNA (RAPD) technique. The study has indicated that the screened primer S2032 can make the green cotton of Xincai 4 produce peculiar amplification from the experiment materials of the 45-germplasm cottons, and the amplified effect of random primer S2032 is stable and clear. The experimental result proves that the RAPD technique is an effective method in the colored cotton application of the variety identification and seed purity identification.
