中国黄牛卵巢卵母细胞体外成熟与体外受精的研究

摘  要：
自屠宰场和临时屠宰的黄牛体内取卵巢2批共25次,黄牛97头,共得可用卵巢卵母细胞657个。用TCM—199加雌激素(FSH、LH、雌二醇—17β)培养于5％CO_2与95％空气的39℃的CO_2培养箱中,使第1批成熟率达到55.04％,第2批成熟率达到61.7％。第1批受精率达到61.97％,移于兔输卵管中发育到卵裂,移于羊输卵管中发育到晚期桑桩胚和早期囊胚。第2批废卵率为38.3％;第10次的体外培养发育率为23.8％;羊输卵管培养发育率达到91％。利用肝素(0.005mg/ml)使精子获能,授精时的精子浓度为25×10~6/ml。用小滴培养法(小滴量为50μl)加盖无菌石蜡培养。结果证明中国老龄黄牛的卵巢还是有利用价值的。

摘  要：
657 usable ovarian oocytes were collected from 97 Chinese yellow cows slaughtered at the local slaughterhouse in 1986 and 1987. The oocytes obtained were cultured in TCM-199, supplemented with FSH, LH and estrodiol-17B, at an atmosphere of 5％ CO_2 and 95％ air, 39℃ for 24 hours. The matured oocytes were fertilized in vitro with sperm (25×10~8/ml) capacitated by heparin (0.005mg/ml) treatment. The maturation rate for 1986 was 55.04％, and that for 1937 was 61.7％. 61.97％ of the oocytes obtained in 1986 were fertilized and some of them developed into morula or blastula after being transfered to the intermediate recipients (sheep or rabbits). Among the oocytes obtained in 1987, 38.3％ were discarded due to poor quality and 23.8％ developed in vitro to certain embryonic stage, and the developmental rate in the sheep, oviducts reached 91％. The results of this experiment indicated that the ovaries of aged cows can also be good resource of oocytes for in vitro fertilization.
