多触发型人参皂苷Rd结肠靶向制剂处方筛选

摘  要:
以体外释放度作为评价指标,制备并筛选人参皂苷Rd结肠定位包衣片处方。采用高效液相色谱法测定释放度,分别考察崩解剂的种类及用量、壳聚糖用量(20%、30%、40%)、增塑剂种类(PEG6 000、TEC、DEP)、时滞层包衣增重(8%、10%、12%)及pH层包衣增重(8%、10%、12%)对包衣片体外释放度的影响。结果表明,以崩解剂(10%羧甲基纤维素钠∶羧甲基淀粉钠=1∶1)用量10%、壳聚糖用量30%、PEG6 000作为增塑剂、时滞层包衣增重及pH层包衣增重用量均为10%时可获得较为理想的包衣片,达到较好结肠定位效果。

摘  要:
In vitro drug-release rate was determined as the evaluation standard to optimize the formulation of colon-specific coating tablets with Ginsenoside Rd. Method In vitro drug-release rate was determined by HPLC. The type and amount of disintegrating agent,dosage of chitosan(20%,30%,40%),plasticizers(PEG6 000,TEC,DEP),coating weight(8%,10%,12%) with time-dependent pH-dependent materials were investigated respectively. The optimum was ultimately opted as the uniform mixture of equal amounts of 10% carmellose sodium,10% carboxymethylstarch sodium to be 10% as disintegrating agent,dosage of chitosan to be 30%,PEG6 000 as plasticizer,the amount of coating weight(8%,10%,12%) with time-dependent pH-dependent materials to be 10% severally.
