人参皂苷Rd类脂质体制备工艺的优选

摘  要:
为优选人参皂苷Rd类脂质体的制备工艺,评价其质量,采用薄膜分散法制备人参皂苷Rd类脂质体,采用超速-离心滤过法测定包封率,以包封率为评价指标,通过单因素考察和正交试验优化制备工艺,对其质量标准进行了研究.人参皂苷Rd类脂质体的最佳制备工艺为表面活性剂与胆固醇物质的量的比为1∶1,HLB值为4. 3,载药量为20 mg,水合温度为65℃,所制得的人参皂苷Rd类脂质体混悬液包封率为(90. 60±1. 12)%,平均粒径为(691. 00±39. 67) nm,Zeta电位为(-6. 18±0. 36) m V.正交试验优选的人参皂苷Rd类脂质体制备工艺稳定、可行,所得的人参皂苷Rd类脂质体包封率较高、粒径分布均匀、外观形态好.

摘  要:
To optimize the preparation technology of ginsenoside Rd liposomes and evaluate their quality. Ginsenoside Rd liposomes were prepared by thin-film dispersion method,encapsulation efficiency was determined by overspeed-centrifugal filtration,and with encapsulation efficiency as evaluation index,the preparation technology was optimized by single factor investigation method and L9(34) orthogonal test. The qualities of ginsenoside Rd liposomes were also evaluated. The optimized prescription was as follows: the molar ratio of surfactant to cholesterol 1 ∶ 1,HLB value 4. 3,drug loading 20 mg,hydration temperature 65 ℃. The encapsulation rate of ginsenoside Rd liposomes was(90. 60±1. 12) %,the average particle size was(691. 00 ± 39. 67) nm,and Zeta potential was(-6. 18±0. 36) m V. The preparation of Rd liposomes is stable and feasible. The obtained liposomes have higher encapsulation efficiency,small particle size,and good appearance.
