鹿骨胶水解物抗氧化活性及对H_2O_2损伤PC12细胞保护作用的研究

摘  要:
[目的]研究鹿骨胶水解物的抗氧化活性及其对H_2O_2损伤PC12细胞的保护作用。[方法]采用热水抽提法对脱脂脱钙的鹿骨进行蛋白提取,用胃蛋白酶和胰蛋白酶进行双酶酶解得鹿骨胶水解物。以DPPH、·OH自由基清除能力测定鹿骨胶水解物的体外抗氧化活性。利用H_2O_2损伤PC12细胞的模型,探究鹿骨胶水解物(HDBG)对氧化损伤PC12细胞的保护作用。[结果]鹿骨胶水解物具有良好的抗氧化活性,对DPPH自由基和·OH均有一定的清除能力,IC_(50)值分别为10.05和4.76 mg/mL。鹿骨胶水解物对正常PC12具有明显的增殖作用,对H_2O_2诱导损伤的PC12细胞具有显著的保护作用,且呈剂量依赖关系。在250、750和1 500μg/mL时,细胞活力分别53.79%、73.16%和82.43%。[结论]鹿骨胶水解物具有良好的抗氧化活性,对H_2O_2损伤PC12细胞具有保护作用。

摘  要:
[Objective] The antioxidative activity of deer bone gel hydrolysate(HDBG) and its protective effect on PC12 cells damaged by H_2O_2 were studied.[Method]Defatted and decalcified deer bones were extracted by hot water extraction and digested with pepsin and trypsin to get glue hydrolysate of deer bone.The anti-oxidation activity of glue hydrolysate of deer bone was measured by DPPH,·OH radical scavenging ability.In vivo,the antioxidant activities of hydrolysate of deer bone glue(HDBG) were investigated by using oxidative injury protection were assessed by using PC12 oxidative injury model induced with H_2O_2.[Result]HDBG had good antioxidant activity and had certain scavenging ability to DPPH free radicals and ·OH.The IC_(50) values were 10.05 and 4.76 mg/mL,respectively.HDBG had a significant proliferation effect on normal PC12 cell,and protected which treated with H_2O_2 in a dose-dependent manner,significantly.At the concentration of 250,750 and 1 500 μg/mL,the cell viability was 53.79%,73.16% and 82.43%,respectively.[Conclusion] HDBG has good antioxidant activity and has protective effect on H_2O_2 damage PC12 cells.
