印度南瓜转录组SSR信息分析及其多态性研究

摘  要:
对印度南瓜(Cucurbita maxima)开展转录组测序分析,共获得131 960条unigene(90.80 Mb),筛选得到12 557个SSR位点(占总unigene的9.52%),其发生频率为1/7.4 kb;其中SSR位点中主导类型为二核苷酸重复,占总SSR的49.82%;其次是三核苷酸重复,其出现频率为45.31%。通过Primer5设计得到7 290对SSR引物,随机选择20对SSR引物对30种不同来源的南瓜进行多态性验证分析,结果显示在14对可扩增产物的SSR引物中,11对引物表现稳定可重复的多态性,UPGMA多态性分析显示11对引物可将30份南瓜材料分为5类。研究表明通过对南瓜转录组分析可获得较高频率的SSR位点且类型丰富,为印度南瓜遗传多样性分析和遗传图谱构建提供了候选标记。

摘  要:
A total of 131 960 unigenes(90.80 Mb)were obtained by using transcriptome sequenceing analysis in Cucurbita maxima,and 12 557 simple sequence repeat(SSR)loci(9.52%)with the frequency of 1/7.4 kb were then identified. Among all SSR loci,dinucleotide repeat was the main type(49.82%),followed by trinucleotide repeat motif(45.31%). A total of 7 290 SSR primers were designed and 20 primers were selected randomly for PCR amplification using 30 pumpkin materials with different sources. Results showed that 14 primers could produce clear and reproductive bands,within which 11 SSR markers performed polymorphical bands. The pumpkin materials could be divided into five groups according to the UPGMA analysis. Results of this present study indicated that SSR markers with high frequency and various types could be acquired by transcriptome sequencing. The SSR markers obtained in the present study could be candidate markers for genetic diversity analysis and genetic mapping construction in Cucurbita maxima.
