不同光周期对肉鸡免疫器官褪黑激素受体表达的影响

摘  要:
为研究不同光周期对肉鸡胸腺和脾脏褪黑激素受体(Mel1a、Mel1b、Mel1c)表达的影响,阐明光周期影响肉鸡免疫功能的可能途径,本试验选用0日龄雄性817肉鸡45只,按照光照∶黑暗为8 h∶16 h、12 h∶12 h、16 h∶8 h三种不同的光周期随机分为Ⅰ、Ⅱ、Ⅲ组。饲养2周后取肉鸡胸腺和脾脏组织,测定其器官指数,采用反转录聚合酶链式反应(RT-PCR)方法检测其褪黑激素受体Mel1a、Mel1b和Mel1c基因的相对表达水平。结果显示,与Ⅱ组和Ⅲ组相比,Ⅰ组胸腺指数分别提高了16.13%(P<0.05)和22.03%(P<0.05),脾脏指数分别提高了19.40%(P>0.05)和21.21%(P<0.05),胸腺Mel1a基因相对表达水平分别升高了27.25%(P<0.05)和88.56%(P<0.05),脾脏Mel1a基因相对表达水平分别升高了11.53%(P<0.05)和19.08%(P<0.05),胸腺Mel1b基因相对表达水平分别升高了15.59%(P>0.05)和30.65%(P<0.05),脾脏Mel1b基因相对表达水平分别升高了18.07%(P>0.05)和40.11%(P<0.05),胸腺Mel1c基因相对表达水平分别升高了17.79%(P<0.05)和70.14%(P<0.05),脾脏Mel1c基因相对表达水平分别升高了34.05%(P<0.05)和63.77%(P<0.05)。结果表明,短光照可提高肉鸡免疫器官指数和褪黑激素受体基因的相对表达水平。

摘  要:
To investigate the effects of different photoperiods on the expression of melatonin receptors(Mel1a, Mel1b, Mel1c) in thymus and spleen of broilers, and elucidate the possible pathways through which photoperiods influence the immune function of broilers, this study selected 45 male 817 broilers at 0-day-old and randomly divided them into three groups(Group Ⅰ, Group Ⅱ, Group Ⅲ) according to the photoperiods of 8 h light∶16 h dark, 12 h light∶12 h dark and 16 h light∶8 h dark, respectively. After two weeks of feeding, the thymus and spleen tissue were collected, and the organ indexes were determined. The relative expression levels of Mel1a, Mel1b and Mel1c genes were detected by reverse transcription-polymerase chain reaction(RT-PCR). The results showed that compared with Group Ⅱ and Group Ⅲ, the thymus index of Group Ⅰ was increased by 16.13%(P<0.05) and 22.03%(P<0.05), and the spleen index increased by 19.40%(P>0.05) and 21.21%(P<0.05), respectively. The relative expression levels of Mel1a gene in the thymus increased by 27.25%(P<0.05) and 88.56%(P<0.05), and in the spleen increased by 11.53%(P<0.05) and 19.08%(P<0.05) in Group Ⅰ compared with Group Ⅱ and Group Ⅲ, respectively. The relative expression levels of Mel1b gene in the thymus increased by 15. 59%( P > 0. 05) and 30. 65%( P < 0. 05),and in the spleen increased by 18. 07%( P > 0. 05) and 40. 11%( P < 0. 05) in Group Ⅰ compared with Group Ⅱ and Group Ⅲ,respectively. The relative expression levels of Mel1c gene in the thymus increased by 17. 79%( P < 0. 05) and 70. 14%( P <0. 05),and in the spleen increased by 34. 05%( P < 0. 05) and 63. 77%( P < 0. 05) in Group Ⅰ compared with Group Ⅱ and Group Ⅲ,respectively. The results indicate that short photoperiods can increase the immune organ indexes and relative expression levels of melatonin receptor genes of broilers.
