摘  要:
本文利用PCR技术扩增得到香蕉束顶病毒(BBTV)NSP株系DNA组分5的全基因,该基因全长为1 013 nt,具有一个开放阅读框,编码161个氨基酸。NSP株系与南太平洋组澳大利亚、夏威夷、埃及分离物DNA组分5核苷酸和编码的氨基酸序列相比较,核苷酸序列同源率介于88%~89%之间,氨基酸序列同源率介于76%~79%之间,借助进化树分析和生物信息学研究方法,发现NSP株系组分5同样含有保守的与植物体内Rb蛋白结合的、能够调节寄主体内DNA复制相关蛋白积累的功能基序———LYCDE;并预测了蛋白质二级结构和性质。

摘  要:
The DNA component 5 (DNA-5) of Banana bunchy top virus (BBTV) NSP strain was cloned and sequenced. It was 1 013 nts in length and could code for 161 amino acids. The nucleotide and amino acid sequences of the DNA-5 were compared with those of isolates from Australia, Hawaii, Egypt, and identities of nucleotide and amino acid sequence among these isolates were between 88%- 89% and 76%- 79%respectively. Phylogenetic and bioinformatics analysis showed that there was a conserved functional motif-LYCDE in DNA-5, which could bind to Rb protein in plant to adjust the accumulation of some proteins related to plant DNA replication. In addition, the secondary structure and some physicochemical properties of the DNA-5 protein were predicted.
