干扰素诱导蛋白204在结肠炎发生中的作用

摘  要:
干扰素诱导蛋白204(IFI204)作为胞内DNA受体,在限制DNA病毒复制和胞内菌感染中发挥重要作用。然而,IFI204及其所介导的信号的活化在肠道中的作用仍不清楚。为了探究IFI204在结肠炎发生中的作用,用葡聚糖硫酸钠(DSS)诱导小鼠结肠炎,通过对结肠长度的变化、临床病理损伤、组织病理损伤以及中性粒细胞浸润情况进行评价,确认DSS诱导的结肠炎的发生。随后,通过定量PCR方法确定IFI204在各个组织中的表达情况,进一步用Western blot方法检测IFI204蛋白在结肠组织中的表达情况,确定结肠组织中确实有IFI204表达。研究发现,IFI204在结肠中有表达,DSS诱导肠炎后,结肠组织中IFI204蛋白的表达降低,其下游信号干扰素基因刺激蛋白(STING)的表达及其磷酸化水平均显著降低,表明IFI204在结肠中具有保护作用。

摘  要:
IFI204 is an intracellular DNA receptor,and it plays an important role in limiting DNA virus replication and intracellular bacterial infection. However,the role of IFI204 and its mediated signal activation in the gut remains unclear. To explore the role of IFI204 in colitis,we first established a DSS model,and then evaluated changes in colon length,clinic-pathological lesions,histopathological lesions and neutrophil infiltration so as to confirm that DSS-induced colitis was successfully established. Subsequently,we used the quantitative PCR method determine the expression of IFI204 in various tissues; and we did observe IFI204 expression in colon tissue. Furthermore,we used Western blot to examine IFI204 expression in colon and found that after DSS-induced enteritis,this expression was decreased,and the downstream signal STING protein expression and the phosphorylation level were significantly reduced. In summary,IFI204 was expressed in the colon,when in colitis,the expression of IFI204,its downstream signaling molecule STING protein were decreased,which indicated that IFI204 had a protective effect in colon.
