采用SPE-HPLC法检测分析桑树喷施阿苯达唑后药液在叶片中的持留量及消解规律

摘  要:
将苯并咪唑类药物阿苯达唑喷施桑树后采叶喂蚕可预防和治疗家蚕微粒子病。准确测定与分析阿苯达唑在桑叶中的持留量及消解规律,对明确药物时效性以及确定施药方案十分重要。建立了固相萃取-高效液相色谱(SPE-HPLC)联用技术测定桑叶中阿苯达唑持留量的方法:桑叶样品中的药物成分用乙腈提取,经固相萃取氨基柱净化后,用高效液相色谱分离,再用二极管阵列检测器(DAD)检测以及外标法定量。方法学验证结果表明:药物浓度检测标准曲线相关系数R2=0.999 1;日内和日间的精密度良好,色谱峰保留时间的变异系数(RSD)为1.33%~3.88%,峰面积的RSD为2.40%~5.69%;样品回收率在80.67%~108.30%;最低检测限(LOD)和最低定量限(LOQ)分别为2.43 ng/L、8.09 ng/L。利用该方法对2次试验中分别以1 000 mg/L和1 500 mg/L阿苯达唑悬浊液喷施后的桑叶样品进行检测分析,各浓度药液处理区桑叶中的药物消解规律均符合一级动力学方程,半衰期在2.22~3.22 d。该检测方法具有准确、灵敏、快速的特点,可应用于桑叶中阿苯达唑持留量及消解规律的检测分析。

摘  要:
Albendazole( ABZ),a benzimidazole parasiticide,could be used to prevent and control silkworm( Bombyx mori) pebrine disease after spraying onto mulberry leaves and feeding silkworms. Accurate determination and analyses of retention and degradation dynamics of ABZ in mulberry leaves plays a critical role in determining drug timeliness and application scheme. Determination method of ABZ retention in mulberry leaves was established by solid phase extraction and high performance liquid chromatography( SPE-HPLC). ABZ in mulberry leaf sample was extracted with acetonitrile first,and then purified by Cleanert NH2solid-phase extractor. After being separated by HPLC and detected by Diode Array Detector( DAD),ABZ content was determined by external standard method. Results of methodology validation suggested that correlation coefficient of standard curve for ABZ content determination R2 was 0. 999 1,intra-day and inter-day precision were excellent,RSD of retention time was within the range of 1. 33% to 3. 88%,RSDs of peak area was from 2. 40% to 5. 69%,the recovery rate of sample was from 80. 67% to 108. 30%,and the minimum detection limit and quantitation limit was 2. 43 ng / L and 8. 09 ng / L respectively. ABZ content in mulberry leaf samples was determined after spraying ABZ suspension of 1 000 mg / L and 1 500 mg / L respectively in two tests. The result indi-cated that the degradation dynamics of ABZ in mulberry leaves from different concentrations of treatment groups was consistent with the first order kinetic equation,and its half-life period was 2. 21 to 3. 22 d. This detection method is accurate,sensitive and speedy and could be used to determine and analyse retention and degradation dynamics of ABZ in mulberry leaves.
