牛乳铁蛋白抗体的制备及其特性测定

摘  要：
利用BLF抗原免疫BALB/c小鼠,采用常规细胞融合技术制备了6株抗BLF的单克隆抗体细胞株,并用此抗原免疫新西兰大白兔获得了55 m L的兔多抗血清。针对腹水单抗及多抗血清进行了一系列特性分析,结果显示,5株抗体效价基本上在1∶10~6以上;间接ELISA测定6株抗体与酪蛋白、乳白蛋白和β-乳球蛋白基本无交叉反应,抗体特异性良好;抗体为IgG1亚类,2B12抗体亲和力优于其他抗体。多抗血清效价可达到1∶10~7以上,抗体也具有一定的特异性。

摘  要：
Six strains of monoclonal antibody(McAb) cells against bovine lactoferrin(BLF) were obtained with BLF antigen immune BALB/c mice by the hybridoma cells technique, and the six McAbs were detected on the antibody titer, specificities and relative affinity; the BLF antigen was used with injection to immunize the New Zealand white rabbit so as to get 55 m L of rabbit polyclonal antibody(Pc Ab) serum; and the ascites fluid McAb and PcAb sera were analyzed on a series of characteristics in this study. The results showed that the titer of five of the six Mc Abs was basically over 1 ∶ 106; the six Mc Abs with good antibody specificity were of almost no cross reactivities with other proteins in milk, such as casein, lactalbumin and β-lactoglobulin, by the indirect ELISA; and the antibody in IgG1 subclass was, of higher 2B12 antibody affinity than other antibodies. The Pc Ab serum could be of the titer of over 1 ∶ 107, and the Pc Ab was of a certain specificity, i.e. low reactivities with casein, lactalbumin and β-lactoglobulin.
