乳铁蛋白酶标抗体的制备及其酶联免疫检测方法的建立

摘  要:
采用过碘酸钠法对抗牛乳铁蛋白抗体进行了辣根过氧化物酶标记,建立了二步式的双抗体夹心ELISA法,并对市售奶粉样品进行了测定。结果表明:所制备的酶标记抗体效价达到了1.6×106,与乳中主要其他蛋白如酪蛋白、乳白蛋白和β-乳球蛋白基本没有交叉反应,特异性较好;将HRP-2B12稀释105倍,以1%OVA为封闭液、含3%BSA的PBST溶液为酶标抗体稀释液,在此条件下建立的ELISA方法实现了对奶粉样品的定性测定。

摘  要:
The anti-bovine lactoferrin antibody was labeled by sodium periodate oxidation method, and a sandwich ELISA system against bovine lactoferrin was developed for determining bovine lactoferrin in infant formula milk power. The results showed that the titer of labeled antibody was over 1:1.6×106, and its specificity was of almost no cross reactivities with other proteins in milk, such as casein, lactalbumin and β-lactoglobulin. Diluting HRP-2B12 105 times, with 1% OVA as sealing fluid, PBST solution containing 3% BSA as enzyme mark antibody, the bovine lactoferrin in infant formula milk power was qualitatively detected by the sandwich ELISA system.
