不同牛种GH基因外显子5序列变异研究

摘  要:
在测定牛亚科3个牛种(雷琼牛、巴州牦牛和巴州蒙古牛)GH基因外显子5序列的基础上,分析了不同牛种的序列变异特点,同时引用其它牛种(普通牛、瘤牛、牦牛和水牛)的序列构建分子系统树以探讨牛亚科家畜的系统发生关系。结果表明,在雷琼牛、巴州牦牛和巴州蒙古牛中共检测到5个变异位点,核苷酸取代方式仅有转换和颠换,核苷酸平均突变率仅为1.66%,说明这3个牛种GH基因外显子5多态性较为贫乏;5个变异位点中仅有1个为错义突变,导致亮氨酸和缬氨酸的转换,但该突变在牛种中分布有差异。从构建的系统树来看,普通牛与瘤牛先聚为一类,然后再与牦牛聚在一起,最后才与水牛聚在一起,与已有的分类结果大体一致。本研究的结果支持中国南方可能是一支瘤牛发源地的推论,也支持中国牦牛在起源进化历程中可能与瘤牛发生了基因交流的观点。

摘  要:
Characters of sequence variation at exon 5 of GH(growth hormone) gene of three bovine species(Leiqiong cattle,Yak and Mongolia cattle from Xinjiang Autonomous Region) were analyzed based on sequencing.Together with the cited sequences of other species(cattle,zebu cattle and buffalo),the molecular phylogenetic tree was therefore built to probe phylogenetic relationship between Bovine subfamilies.Total 5 variation sites were examined and two types of mutation,transition and transversion,were observed,with the average percentage of nucleotide variation of 1.66%.It demonstrated the poverty of polymorphism at exon 5 of GH gene of three bovine species. There was only one missense mutation out of 5 mutation sites which led to the shift between leu and val amino.Bos taurus(cattle) was clusterd with Bos indicus(zebu cattle) firstly,then clustered with Bos grunniens(yak) and with Bubalus bubalus(buffalo) lastly,which was in accordance with prior classification.The results were in favour of the opinions that Southern China might be a cradle of Bos indicus and that gene intercourse might have occured between Bos grunniens and Bos indicus in the process of evolution.
