甘草酸微生物转化菌的筛选鉴定及发酵条件优化

摘  要:
首先,采用单因素方法对碳源、氮源、发酵接种量、初始p H、发酵时间、发酵温度等因素进行考查,在此基础上进行4因素3水平的正交试验优化。得到的优化发酵条件是甘草浸出液10%,发酵温度40℃,p H 5.0,接种量10%,装液量75 m L/250 m L,发酵时间为6.5 d,180 r/min。在最优条件下,甘草次酸的量达到7.67 mg/m L。

摘  要:
Firstly,using single-factor test,the effects of these 5 parameters,i.e.,carbon source,nitrogen source,the inoculation amount,initial p H of medium, fermentation time,fermentation temperature on the glycyrrhetic Acid yield were studied. Then the key parameters of fermentation conditions were statistically optimized through the orthogonal array experiments with 4 factors in 3 levels. The optimal conditions were licorice digester liquid 10%,initial p H 5.0,rotation speed 180 r/min,10% inoculum size in 75 mL working volume(in 250 mL flasks) at 40 ℃ for 6.5 d. Using these conditions,the the glycyrrhetic acid yield was 7.67 mg/mL.
