西藏小型猪与军牧一号白猪1GFBP-6基因表达差异分析

摘  要:
采用实时荧光定量PCR(RealTimePCR)方法,对不同生长阶段的西藏小型猪和军牧一号白猪的肝脏、脾脏、肾脏及肌肉组织中胰岛素样生长因子结合蛋白6(IGfBP-6)基因mRNA表达水平进行检测,并利用t检验,对相同生长阶段的各组织2个品种猪之间的表达水平进行比较分析.结果表明:西藏小型猪与军牧一号白猪IGFBP-6基因均在肌肉组织中表达量最高,而在肝组织中表达量最低.IGFBP-6基因在1日龄肾脏中,西藏小型猪表达水平高于军牧一号白猪,差异显著(P〈0.05);在脾脏组织中,西藏小型猪表达水平低于军牧一号白猪,表达水平差异极显著(P〈0.01).其他生长阶段和组织中,2个品种猪IGFBP-6基因表达水平差异均不显著(P〉0.05).

摘  要:
Insulin-like growth factor binding protein 6 (IGFBP-6), a member of insulin-like growth factor binding protein family (IGFBPs), was given extensive attention by researchers because it has high affinity with IGF-2 compared with IGF-1. However, the function of IGFBP-6 gene in pig growth and development is unclear. In this research, the mRNA expression levels of IGFBP-6 were tested by real time PCR in liv- er, kidney, spleen and muscle of Tibetan mini-pig and Junmu No. 1 White pig, and T test was used to analyze the expression levels between the two breeds at the same growth stage. The result showed that IGFBP-6 gene was expressed widely in pigs. The expression level of IGFBP-6 gene in muscle was the highest, while in the liver it was the lowest. IGFBP-6 mRNA expression level in kidney was significantly different between Tibetan mini-pigs (ld) and Junmu No. 1 White pigs (1 d) (P 〈0.05). The expres- sion level in spleen was extremely significantly lower in Tibetan mini-pigs ( 1 d) than Junmu No. 1 White pigs( 1 d) (P 〈 0.01). But IGFBP-6 mRNA expression in other tissues showed no significant difference among different breeds. This research will support function study of IGFBP-6 gene at the molecular level, and the results may provide useful data for further basic research to explore the mechanism of dwarfism in these pigs.
