广西巴马小型猪脂联素cDNA的克隆及序列分析

摘  要:
为了克隆广西巴马小型猪脂联素基因的cDNA全序列及序列分析,试验利用引物悬挂延伸PCR法,以基因组总DNA为模板,先分别扩增得到编码脂联素的第2外显子和第3外显子的序列,再以这2个外显子为模板,进行第2轮PCR,纯化连接pMD 18-T载体后转化大肠杆菌DH5α,筛选阳性克隆体,鉴定后测序。结果表明:经PCR产物测序证明,得到大小与脂联素基因编码序列大小相同的片段,此片段长度为732 bp;同源性比较分析发现,广西巴马小型猪脂联素cDNA序列与GenBank中已报道的家猪脂联素同源性高达99.7%,有2处发生了碱基错义突变。

摘  要:
To clone and analyze the sequence of adiponectin cDNA of Guangxi Bama mini-pig,the sequence of adiponectin cDNA was amplified by over-bang extension PCR protocol using Guangxi Bama genome as template and then ligated into pMD18-T vector after purification.The recombined pMD18-T vector was transformed into the coliform DH5α for sequencing.The cloned adiponectin cDNA gene from Guangxi Bama mini-pig contained 732 bp.The homology analysis showed that compared with the sequence of other pig breeds reported in GenBank,the homology rate of the adiponectin cDNA sequence were 99.7%.The result indicated that the sequence of adiponectin cDNA of Guangxi Bama mini-pig was successfully cloned in the study.
