油茶种质倍性的SSR-PAGE和SSRseq鉴定

摘  要:
本研究采用流式细胞仪、SSR-PAGE和SSRseq相结合的方法,对97份油茶种质资源的染色体倍性进行了鉴定。结果表明:(1)以二倍体狭叶油茶为对照,流式细胞仪检测发现,在96份油茶种质中,29份是二倍体,58份是四倍体、9份是六倍体;(2)每份种质的56个SSR标记的SSR-PAGE扩增位点等位基因数表明,等位基因数最大为2的二倍体有22个,最大为4的四倍体有66个,最大为6的六倍体有9个,流式细胞仪检测的8个二倍体被校正为四倍体;(3)每份种质的56个SSR标记的SSRseq等位基因数鉴定的96份油茶种质染色体倍性结果与SSR-PAGE扩增位点等位基因数估测的结果一致。流式细胞仪可快速检测植物DNA分子量、SSR-PAGE可提供位点的最大等位基因数、SSRseq可精确获得等位基因数,三者结合是快速准确鉴定多倍性物种的染色体倍性的可靠技术与方法。

摘  要:
Chromosome ploidy of 97 germplasms of Camellia oleifera were determined by the methods of flow cytometry, SSR-PAGE(polyacrylamide gel electrophoresis) and SSR sequencing. Results showed that taking diploid C. fluviatilis var. megalantha as the control sample, 29 diploids, 58 tetraploids and 9 hexaploids were identified from 96 samples by analysis of flow cytometry. The number of alleles of SSR-PAGE amplification loci of 56 SSR markers in each germplasm indicated that there were 22 diploid alleles with the maximum number of alleles of 2,66 of the tetraploid with the maximum of 4, and 9 of the six diploid with the maximum of 6. The 8 diploid detected by flow cytometry were corrected to tetraploid. The results of chromosome ploidy of 96 Camellia oleifera germplasms identified by 56 SSR labeled SSRseq alleles per germplasm were in agreement with the results of the number of alleles of SSR-PAGE amplification loci. The flow cytometry could quickly determine DNA molecular weight of plants, SSR-PAGE could provide allele number of amplified loci information, and SSRseq could accurately present number of alleles. The combination of these three methods could provide a quick and reliable technique and method for the identification of chromosome ploidy level of polyploidy species.
