两株细菌纤维素产生菌发酵条件优化初探

摘  要:
【目的】对分离获得的二株菌巴氏醋杆菌(ZHC菌)和葡萄糖酸杆菌(TAⅢ菌)的纤维素发酵工艺进行优化研究。【方法】采用单因素实验法和正交试验法,对两株菌进行发酵条件的初步优化,并对优化后的培养基进行产细菌纤维素的验证实验。【结果】单因素碳源实验表明,两株菌都能较好利用的碳源有葡萄糖、甘油、D-果糖、蔗糖,其中以甘油效果最佳;单因素氮源实验表明,两株菌对硫酸铵的利用效果都较差,蛋白胨和酵母膏为最佳复合氮源;验证实验表明:ZHC菌以培养基:甘油3%、蛋白胨1%、酵母膏0.45%、磷酸氢二钠0.2%,30℃,接种量5%,静置发酵10 d时,细菌纤维素产量达4.24 g/L。TAⅢ菌以培养基:甘油4%、蛋白胨1.2%、酵母膏0.45%、磷酸氢二钠0.3%,30℃,接种量5%,静置发酵10 d时,细菌纤维素产量达3 g/L。【结论】在利用巴氏醋杆菌(ZHC菌)和葡萄糖酸杆菌(TAⅢ菌),发酵生产细菌纤维素时,甘油是最佳碳源选择,蛋白胨和酵母膏为最佳氮源选择。

摘  要:
【Objective】To study the fermentation conditions optimization of two screened strains Acetobacter pasteurianus ZHC and Gluconobacter oxydans TAⅢ. 【Method】Use the method of single factor experiment and orthogonal experi ment method,Preli minary. 【Result】The experimental results show that,Both of the ZHC and TAⅢ the best carbon source the same for glycerol,The best nitrogen source was the same peptone and yeast extract complex nitrogen source. Optimized medium for ZHC bacteria is Glycerol 3% Peptone1% Yeast extract 0. 45% disodium hydrogen phosphate 0. 2%. 30 degrees,Inoculation of 5% ZHC bacteria,Static fer mentation for 10 days,Bacterial cellulose production AMounted to 4. 24 g / L. TAⅢ bacteria is Glycerol 4% Peptone 1. 2% Yeast extract 0. 45% disodium hydrogen phosphate 0. 3%. 30 degrees,Inoculation of5% TAⅢ bacteria,Static fer mentation for 10 days,Bacterial cellulose production amounted to 3 g / L.
