绞股蓝多糖对地塞米松诱导小鼠胸腺和脾脏凋亡影响的研究

摘  要:
本研究采用光学显微镜,透射电子显微镜,琼脂糖凝胶电泳,原位末端标记(TUNEL),过氧化物酶标记的链霉卵白素染色试剂盒(SP)等技术,对腹腔注射地塞米松诱导小鼠胸腺、脾脏淋巴细胞凋亡的模型下,观察绞股蓝多糖对小鼠胸腺、脾脏淋巴细胞凋亡的影响,结果发现,凋亡组和多糖组可见明显的细胞凋亡的特征,胸腺和脾脏淋巴细胞胞核染色质固缩凝聚,有的形成凋亡小体。琼脂糖凝胶电泳显示,凋亡对照组和绞股蓝多糖组呈现典型的梯状条带。统计分析表明,地塞米松诱导凋亡的小鼠试验中,各浓度的绞股蓝多糖对脾脏和胸腺淋巴细胞的凋亡都有保护作用,并随着浓度的增加凋亡细胞减少,而各组间的凋亡细胞数差异极显著(P<0.01)。

摘  要:
This study was carried out using the optical microscope,transmission electron microscope,agarose gel electrophoresis(AGE),TUNEL,SP to detect the effect of gynostemma polysaccharide on lymphocyte apoptosis in the spleen and the thymus of mouse through the hexadecadrol induced apoptosis model.The results showed that the feature of apoptosis was found in apoptosis group and the polysaccharide group in apoptosis model,chromatin pyknosis and cohesion of the spleen and the thymus lymphocyte cell nucleus,some of which can be formed the apoptotic body.AGE showed DNA ladder at the apoptosis group and the gynostemma group.The result from statistical analysis indicate that every concentration of gynostemma can protect the cell from apoptosis in spleen and the thymus the number of the apoptotic cell decreased with the increasing concentration,which was significantly different(P<0.01).
