摘  要:
为探讨辣蓼黄酮正丁醇部分(FNB)对内毒素血症小鼠炎性细胞因子的影响,采用酶解-超声偶联法提取辣蓼全草中的总黄酮,结合大孔树脂纯化富集获得辣蓼正丁醇部分黄酮,通过不同剂量的FNB作用于脂多糖(LPS)刺激所诱导的小鼠内毒素血症模型,测定丙二醛(MDA)、MPO、T-AOC、T-SOD、GSHPX、GSH、LZM、ACP及炎性细胞因子肿瘤坏死因子α(TNF-α)、干扰素(IFN,IFN-α、IFN-γ)、白介素-2(IL-2)水平。结果表明,FNB能通过提升小鼠肝脏的抗氧化防御酶促体系的活性,减少LPS刺激所诱导的促炎因子TNF-α、IL-1β、IL-6、IL-8的释放,降低肺部TNF-α、IFN-α、IFN-γ以及IL-2mRNA的表达水平。说明一定浓度的FNB能减轻LPS诱导的内毒素血症对小鼠的损伤,改善生存率。

摘  要:
In order to investigate the effect of FNB from Polygonum hydropiper L.on inflammatory cytokines in mice with endotoxemia,total flavonoids in the whole plant of Polygonum hydropiperum were extracted by enzymolysis-ultrasonic coupling method.The FNB part of the flavonoids were obtained by extracting and separating,followed with macroporous resin purification and enrichment.The different doses of FNB were used to treat endotoxemia induced by lipopolysaccharide(LPS)in mice.The levels of MDA,MPO,T-AOC,T-SOD,GSH-PX,GSH,LZM and ACP and inflammatory cytokines TNF-α,IFN-α,IFN-γ,IL-2 were determined.The results showed that FNB enhanced the activities of antioxidant defense enzymatic system.FNB can reduce the release of pro-inflammatory factors TNF-α,IL-1β,IL-6 and IL-8 induced by LPS,and the expressions of TNF-α,IFN-α,IFN-γ,IL-2 mRNA.In brief,FNB attenuated the injury of endotoxemia and improved the survival rate in mice.
