山羊卵巢卵母细胞的体外受精试验

摘  要：
将18枚细胞质均匀、包有完整致密卵丘细胞的山羊小卵泡(2～5mm)卵母细胞培养于含有hCG(20μg/mL)和FCS(10％)的TCM_(199)中30h,然后用经含有肝素(20μg/mL)和BSA(10mg/mL)的BO液诱导获能的山羊新鲜精子进行授精处理,授精后8h,转移入含有10％FCS的TCM_(199)中继续培养12h,结果有10枚卵子成熟,成熟率为55.6％,7枚卵子受精,受精率为70％。把7枚原核期受精卵移植给3只同期发情的受体山羊,2只妊娠,妊娠率为66.7％,并获得一只雄性试管山羊胎羔,胎龄为52d。

摘  要：
Immature oocytes were collected from 2 to 5 mm diameter follicles of goat ovaries obtained surgically. Eighteen oocytes with the compact and dense cumulus cell and homogeneous ooplasm were incubated in TCM199 (Earle's salts) supplemented with hCG(20 μg / mL) and FCS(10%) under sterile paraffin oil at the conditions of 39 and 5% CO2 in air for 30 h. Of 18 oocytes, 10 released first polar bodies. The maturation rate was 55.6%. Fresh goat semen were washed twice through centrifugation (1500 r / min, 10 min) with BO medium without BSA and then incubated in BO medium with BSA (10 mg / mL) and heparin (20 jμg / mL) at 39 for 2 h. After insemination for 8 h, the eggs were cultured in TCM,99 with FCS(10%) for 12 h. 7 of 10 eggs (70%) were fertilized. The 7 fertilized eggs (in pronuclear stage) were transferred into 3 synchronized recipients. 2 were pregnant. One of the pregnant recipients died from injuries at 52 day after transfer. One developmentally normal fetus, 8.5 cm long was obtained. Mammary gland well development was found from the other recipient in two monthes. However, no kid was born.
