体外培养前黄牛卵母细胞的分类、形态结构及成熟能力的研究

摘  要:
自1-5mm卵泡中负取的卵母细胞在实体显微镜下。按形态可分为四类:(1)外观正常的;(2)少丘带斑的;(3)扩展后退化的;(4)少丘均质的。通过中性红/台盼蓝染色、二乙酰荧光素染色,电镜观察和体外成熟实验等方法系统地研究了这四类卵母细胞的形态和发育能力,为牛卵母细胞体外成熟和体外受精提供了基础数据。研究结果说明第一类卵母细胞处于最佳生理状态,体外成熟率最高;其它三类卵母细胞生理状态欠佳,成熟率很低。

摘  要:
The experiment was designed to evaidate the iiving state and developmental ability of different types of follicular ootytes from the Chinese yellow cowsws by means of Neat. tral Red/Trypan blue staining, FDA sta(?)ng, electron (?)copy and in vitro (?)tion test, some useful data were obtainedd for the successful in vitro mat(?)tion and fertilization of bovine oocytes. The oocytes aspiraced from (?) folictes on the oya- rian surface were divided into four groups ander the sterecomicroscope.The oocytes in the first group, completely surrounded with compact cumulus oophorus of many layers, made up 35% of the whole population. Those in the second group, either with few cumulus cells or nude and having a speckled cytoplasm, were 32% of the total number of oocytes obtained. The oocytes in the third group, enclosed by expanded cumulus with atretic bodies, accounted for 19% of the total. Although the oocytes in the fourth group were also nude or with few cumulus cells, they were different from the second group in having an evenly granulated cytoplasm. This group mode up 8%. of the total oocyte number. The results of N /T staining showed that oocytes in the first group were in the best living state:in46% of them, neither the cytoplasm nor the cumulus was stained purple, indicating that both were alive. For the test 54% of this group, only the cumulus was stained. Ultrastructurally, the oocytes in the first group were completely normal, with a normal germinal vesicle and organelles of nortmal structure and distribution in the cytoplasm, and almost all of their cumulus cells showed no sign of degeneration. An outstanding feature of the second group oocytes under the EM was that the increased lipid drops and vesicles often clustered and the individual ones were often found surrounded with mitochondria. Most of their cumulus cells degenerated to a big margin. The in vitro maturation test showed that the oocytces in the first group had the highest GVBD rate of 77%; that of the second and fourth group oocytes were the lowest, 30 and 35%, respectively.Although the GVBD rate of the oocytes in the third group appeared to be pretty high (68%),some of themmight have already undergone GVBD before culture.
