维生素C对亚硝酸盐胁迫下匙吻鲟仔鱼非特异性免疫及抗氧化能力的影响

摘  要:
为初步探究水体中添加不同剂量维生素C对匙吻鲟内源营养期及混合营养期仔鱼抗亚硝酸盐胁迫能力的影响,采用生化方法测定不同条件下仔鱼溶菌酶(LZM)、酸性磷酸酶(ACP)、碱性磷酸酶(AKP)活性,丙二醛(MDA)含量和总抗氧化能力(T-AOC)在内源营养期及混合营养期的动态变化。结果显示:以卵黄为营养的内源营养期,LZM、AKP、ACP活性和T-AOC随维生素C质量浓度的增加呈现升高趋势,且90 mg/L和120 mg/L维生素C组显著高于对照组和胁迫组;以卵黄和外源饵料作营养的混合营养期,LZM、AKP、ACP活性随维生素C质量浓度的增加呈现"升高—降低"趋势。90 mg/L维生素C组LZM、AKP、ACP活性和T-AOC从内源营养期到混合营养期显著上升,上升率分别为71.8%、33.7%、48.4%和61.1%。MDA含量随维生素C质量浓度的增加而降低,且在同一取样时间下显著低于胁迫组,维生素C添加组MDA含量从内源营养期到混合营养期无显著变化。综上,在亚硝酸盐胁迫下,90 mg/L维生素C浸泡对匙吻鲟仔鱼内源营养期及混合营养期保护效果较好;亚硝酸盐胁迫可导致匙吻鲟内源营养期及混合营养期仔鱼机体的氧化损伤及抗氧化能力的不平衡,但不同质量浓度维生素C浸泡可以在一定程度上缓解亚硝酸盐胁迫的负面影响。

摘  要:
In order to investigate the effects of different concentrations vitamin C( Vc) on the non-specific immunity and antioxidant capacity of Polyodon spathula during endogenous and mixed nutrition under nitrite stress,the biochemical methods were employed to determine the activities of lysozyme( LZM),acid phosphatase( ACP),alkaline phosphatase( AKP),and the malondialdehyde( MDA) content and the total antioxidant capacity( T-AOC). The results suggested that during endogenous nutrition,the activities of LZM,AKP,ACP and T-AOC were increased in a concentration dependent manner,and the Vc 90 mg/L and 120 mg/L groups significantly increased( P < 0. 05) compared with the control and the stress groups; during the mixed nutrition,the activities of LZM,AKP and ACP increased first and then decreasedwith the rising of Vc concentration. In Vc 90 mg/L group,the T-AOC and the activities of LZM,AKP,ACP increased by 61. 1%,71. 8%,33. 7% and 48. 4% from endogenous to mixed nutrition. The MDA content was decreased with the rising of Vc concentration,and significantly lower than that of the stress group( P < 0. 05),but no significant differences between endogenous and mixed nutrition stages in all Vc groups. In conclusion,under nitrite stress,90 mg/L Vc soaking could protect the Polyodon spathula larvae during the endogenous and mixed nutrition,and the nitrite stress led to the imbalance of antioxidant capacity and oxidative damage,however,the Vc could alleviate the negative impact of nitrite stress.
