超高效液相色谱内标法测定油菜种子硫苷含量

摘  要:
首次建立超高效液相色谱(UPLC)内标法测定油菜种子中硫代葡萄糖苷(glucosinolate)的方法。以丙烯基硫代葡萄糖苷(sinigrin)为内标,以φ=20%乙腈-水为流动相,采用C18柱(2.1mm×50mm填料1.7μm),流速0.3mL/min,线性梯度洗脱,229nm检测;内标溶液浓度为0.15~2.5mmol/L时,峰面积与浓度线性关系良好,相关系数为r=0.999 53;该方法最低检测限(S/N=3)为1.0μmol/L,最低定量限(S/N=10)为5.0μmol/L,回收率为98.54%~101.71%。该方法测定结果与国标规定的HPLC法测定结果经t检验,无显著性差异;经应用验证,该方法结果准确、快速,能满足油菜种子中硫代葡萄糖苷的测定需求,具有较高的实用价值。

摘  要:
The method was firstly established in rapeseed glucosinolates content determination by ultra performance liquid chromatographic(UPLC).Using sinigrin as internal standard,with 20% acetonitrile and water as mobile phase at a flow rate of 0.3 mL/min.Linear gradient elution with C18 column,and the detection wavelength was set at 229 nm.The correlation coefficient of linear calibration curve was 0.999 53 within the sinigrin concentration range of 0.15-2.5 mmol/L;The limit of detection(S/N=3) was 1.0 μmol/L,and the limit of quantitation(S/N=10) was 5.0 μmol/L.The recovery was found in the range of 98.54%-101.71%.The results showed that it has no significance difference between UPLC and national standard method.The method can meet the demand in determination.
