多杀菌素产生菌株航天育种效果研究

摘  要:
[目的]探索多杀菌素诱变育种的有效途径。[方法]对多杀菌素经"实践八号"育种卫星搭载后的菌株进行稀释涂布分离并对分离后的菌株进行筛选和发酵测定,计算多杀菌素总效价。[结果]搭载后的刺糖多孢菌SP1,经大量筛选得到2株高产且遗传性状相对稳定的菌株HY463和HY466,多杀菌素总效价分别达到147.51μg/ml和95.33μg/ml,产量分别比出发菌株SP1提高288%和151%。对搭载后的菌株进行稀释涂布分离发现,菌落形态各异,有草帽状、中间凹状、放射线状和宝塔状,并且以草帽状的菌落居多,且不同形态的突变菌株发酵水平也参差不齐。[结论]与UV6、0Co等其他诱变方法相比,空间搭载诱变更能使刺糖多孢菌的产素能力得到较大的提高,是选育多杀菌素高产菌株的有效方法。

摘  要:
[Objective] The research was to explore the effective approach for mutation breeding of Spinosad.[Method] Spinosad strains,carried through breeding satellite "Shijian-8",were made for separation by dilution spreading,the separated strains were screened and fermented for determination,and then the total Spinosad titers were calculated.[Result] Two strains HY463 and HY466 with high productivity and stable genetics were obtained through a great deal of screening from carried Saccharopolyspora spinosa SP1.Compared with the original strain SP1,their total Spinosad titers were 147.51 μg/ml and 95.33 μg/ml and yield were increased by 288% and 151% resp.Through the separation on the carried strain by dilution spreading,it was found that the colony had many kinds of types such as strawhat-shaped,concave-shaped,radiationshaped and pagoda-shaped.Among these colonies,the strawhat-shaped colony was more than others and the fermentation level of all the mutants were different.[Conclusion] Compared with other mutation breeding methods such as UV、60Co,the space carrying mutation could more increase the ability of producing Spinosad by S.spinosa,which was effective mode for screening the high yield strains of Spinosad.
