参薯组织培养的多因子正交试验研究

摘  要:
以参薯茎段为研究材料,使用0.1%HgCl2进行不同消毒时间的消毒处理,并应用正交试验设计对参薯无菌苗的诱导、 生根培养进行研究.结果表明:参薯茎段以0.1%HgCl2消毒7 min效果较好,污染率低,存活率及发芽率较高;最佳诱导培养基为:MS+6-BA 0.5 mg/L+GA30.1 mg/L+AC 1.0 g/L+蔗糖30 g/L;最佳生根培养基为:1/2 MS+NAA 0.5 mg/L+IBA 0.5 mg/L+蔗糖25 g/L+AC 0.5 g/L;添加0.5~1 g/L的AC能有效减小参薯组培苗的褐化.

摘  要:
Vine stems of Dioscorea alata were cut into segments and disinfected with 0.1% Hg Cl2 for different durations for microprogation by tissue culture, and an orthogonal experiment was designed and conducted for multiplication and root growth of the disinfected stem segments. The results showed that the stem segments of D. alata was better in disinfection when treated with 0.1% Hg Cl2 for 7 min, and had a lower contamination rate as well as a higher survival rate and a sprouting rate. The best medium for inducing and multiplication was MS+6-BA 0.5 mg/L+GA30.1 mg/L+AC 1.0 g/L+sucrose 30 g/L; the best rooting medium was 1/2 MS+NAA 0.5 mg/L+IBA 0.5 mg/L+sucrose 25 g/L+AC 0.5 g/L; supplement of0.5~1 g/L of AC in the medium could effectively reduce the browning of the tissue culture plants.
