响应面法优化黑曲霉发酵产单宁酶条件

摘  要:
通过单因素试验、中心组合实验设计及响应面法对黑曲霉的产单宁酶条件进行优化。确定对发酵产酶影响最大的因素依次为:单宁浓度、培养温度、培养时间。其最适产酶条件为:31.7℃,转速180r/min,单宁浓度1.5%,初始pH 6.0,接种量10%.,装液量20%,培养时间72h。在此条件下,发酵单宁酶活力可达1 141.82U/mL。

摘  要:
Through single factor experiments,central composite design and response surface method,the conditions of producing tannase of Aspergillus niger were optimized.The most three important factors of determining the tannase production were tannic acid concentration,temperature and incubation time.The optimum conditions for enzyme production are as follows:temperature 31.7℃,speed 180 r/min,tannic acid concentration 1.5%,initial pH 6.0,inoculum 10%,bottle volume 20%,culture time 72 h.With these conditions,the fermentation tannase activity reached 1141.82 U /mL.
