红壤地区不同生态型蒲公英绿原酸含量比较及其抑菌效果

摘  要:
为从多方面探究评价红壤环境对蒲公英绿原酸抑菌效果的影响,为蒲公英在红壤地区的推广种植及以开发绿原酸抑菌剂为主要内容的发展提供理论和试验依据,采集来自红壤不同地区的两种野生型蒲公英作为试验材料,以实验室栽培型蒲公英作为对比材料,通过乙醇回流法,对提取液经减压浓缩后,采用大孔树脂柱层析法分离纯化绿原酸,测定并比较其绿原酸含量差异,并对纯化的绿原酸进行抑菌试验。结果表明:1)不同生态型的蒲公英绿原酸含量纯化前在2.21%~7.03%,纯化后在1.12%~5.44%;2)不同生态型的蒲公英绿原酸在经大孔树脂纯化后平均质量浓度最大增幅可达4100%;3)金华野生型、杭州野生型和种植型3种不同生态型的蒲公英绿原酸对大肠杆菌和金黄色葡萄球菌的抑菌圈从大到小分别依次为11.6mm、10.6mm、8.8mm和10.9mm、9.5mm、8.6mm,对大肠杆菌和金黄色葡萄球菌的最小抑菌浓度从大到小依次为0.040 3 mg/mL、0.021 6 mg/mL、0.005 mg/mL和0.040 3 mg/mL、0.021 6 mg/mL、0.022 1mg/mL。红壤不同地区蒲公英绿原酸含量存在差异,野生型高于栽培型;且纯化后的绿原酸对于试验菌株大肠杆菌和金黄色葡萄球菌均有较好的抑制效果。

摘  要:
In order to provide theoretical and experimental basis for cultivation promotion of dandelion in red soil area and the development,the main content of which is exploiting chlorogenic acid fungistat,and lay the theoretical foundation for exploiting dandelion in red soil area,collecting two kinds of wild dandelion in the different red soil regions as experimental materials,and the dandelion planted in the lab as contrastive materials.Using ethanol return method and decompression enrichment,then separating and purifying chlorogenic acid through column chromatography in macroporous resin. Measuring and comparing the differences of chlorogenic acid contents,and conducting the bacteriostasis experiments.The results showed that the chlorogenic acid contents in dandelion were between 2.21% and 7.03% before purification and between 1.12% and 5.44% after purification in various red soil regions.And the largest increase of average mass concentration of chlorogenic acid of dandelion could be 41.0times in various red soil regions.In the bacteriostatic action,the order from big to small of inhibition zone of chlorogenic acid on Escherichia coli was 11.6mm,10.6mm,8.8mm,and the order from big to small of inhibition zone of chlorogenic acid on Staphylococcus aureus was 10.9mm,9.5mm,8.6mm.The MIC of chlorogenic acid for E.coli and S.aureus were 0.040 3mg/mL,0.021 6mg/mL,0.005 5mg/mL and 0.040 3mg/mL,0.021 6mg/mL,0.022 1 mg/mL.On the whole,there were differences existing in chlorogenic acid of dandelion in various red soil regions,the wild were higher than the planted.And the purified chlorogenic acid had a certain inhibitory effect on experimental strains E.coli and S.aureus.
