摘  要:
以广西柳城县的野生毛葡萄种子作为外植体材料,研究单一灭菌剂、两种灭菌剂组合以及三种灭菌剂组合对种子灭菌效果及种子发芽率的影响,并比较常温无菌水(20℃)、无菌温水(40℃)和添加GA3(0.5mg/L,40℃)的预处理对种子发芽率的影响。试验结果表明:单一灭菌剂0.1%升汞的适宜灭菌时间为16min;两种灭菌剂组合的75%酒精适宜处理时间为30s;三种灭菌剂组合处理灭菌的效果优于两种灭菌剂组合和单一灭菌剂处理,以75%酒精30s+0.1%升汞16min+15%次氯酸钠5min为最佳灭菌组合,野生毛葡萄种子的污染率为16.67%,发芽率为61.67%。三种不同处理中,以GA3(0.5mg/L,40℃)处理的效果最佳,野生毛葡萄种子的发芽率为73.33%。

摘  要:
The seeds of wild vitis quinquangularis in Liucheng County were used as the explant materials and the effects of single sterilization agent, two agents combinations and three agents combinations on the seed sterilization and germinations percentage were studied. And the pretreatment effects of sterile water with common temperature(20℃), sterile water with warm temperature(40℃) and adding GA3(0.5mg/L,40℃) on the seed germination percentage were compared. The experiment results showed that the suitable sterilization time of 0.1%Hg Cl2 was 16min and the time of 75% Alcohol was 30s; the sterilization effect of treatment with three sterilization agents was better than that of single agent and two agents; the best sterilization combination was 75% Alcohol30s+0.1% Hg Cl216min+15% Sodium hypochlorous 5min, in which the pollution rate was16.67% and germination percentage was 61.67%; in the three different pretreatments, the treatment with GA3(0.5mg/L,40℃) showed the best effect and the germination percentage achieved to 73.33%.
