磺胺甲噁唑与吉富罗非鱼血浆蛋白结合率的测定方法研究

摘  要:
为建立磺胺甲噁唑(Sulfamethoxazole,SMZ)与吉富罗非鱼(Oreochromis niloticus)血浆蛋白结合率的测定方法,实验研究了体外平衡透析法测定SMZ血浆蛋白结合率的最佳测定条件,从而推断出体内超滤法的最佳测定条件,最后进行了两种测定方法的对比研究。结果显示:SMZ血浆蛋白结合率不受温度影响,但具有p H依赖性和药物浓度依赖性。在4℃、p H 7. 4条件下,体内外血浆蛋白结合率测定结果如下:0. 5~10. 0μg SMZ/m L透析液范围内,体外SMZ血浆蛋白结合率为15. 48%~25. 12%,10. 0~60. 0μg SMZ/m L透析液内,体外血浆蛋白结合率维持在14. 65%~15. 48%内;在血浆含药浓度为1~5μg/m L内,体内SMZ血浆蛋白结合率为15. 34%~20. 1%;在10~20μg/m L内,体内结合率趋于稳定,维持在12. 7%~14. 9%内,与体外测定结果的变化趋势保持一致。通过两种测定方法的对比研究,确定体内超滤法为SMZ血浆蛋白结合率测定的最佳选择。

摘  要:
The optimal conditions for determination of SMZ plasma protein binding rate by in vitro equilibrium dialysis were studied to establish a method for the determination of plasma protein binding of sulfamethoxazole in Oreochromis niloticus. A comparative study of the two methods was performed. The results showed that plasma protein binding of SMZ was temperature independent,p H dependent and concentration dependent. The results of the in vitro and in vivo plasma protein binding assays at 4 ℃ and p H 7. 4 were tested. In vitro SMZ plasma protein binding rates were ranged from 15. 48% to 25. 12% by dialysate of 0. 5 ~ 10. 0 μg SMZ/m L. In vitro SMZ plasma protein binding rates were ranged from14. 65% to 15. 48% by dialysate of 10. 0 ~ 60. 0 μg SMZ/m L. The plasma protein binding rate of SMZ in vivo was ranged from 15. 34% to 20. 1%and from 12. 7% to 14. 9% by plasma concentration of 1 ~ 5 μg SMZ/m L and 10 to 20 μg SMZ/m L, respectively. Consistent with the trend of changes in vitro measurements. As a conclusion,this experiment determined that the best choice for the determination of SMZ plasma protein binding rate was in vivo ultrafiltration by the comparative study of the two assay methods.
