宣和猪ENHO基因多态性及其与肉质性状的关联分析

摘  要:
为揭示宣和猪能量稳态相关(ENHO)基因多态性及其与肉质性状的关联,采用PCR产物直接测序法对119头宣和猪ENHO基因全序列进行单核苷酸多态性(SNP)位点检测,将各SNP位点基因型及单倍型组合与肉质性状进行关联。结果:在宣和猪ENHO基因上检测出4个SNP变异位点,其中,内含子1区域检测出3个SNP位点(G420A、A896G和A1114G),3′-UTR区检测出1个SNP位点(C1720T)。在4个位点中,除G420A位点呈低度多态(PIC为0.123 9)、遗传多样性较低(H为0.132 7)外,A896G、A1114G和C1720T位点均呈中度多态(PIC介于0.497 1~0.499 7),遗传多样性也较为丰富(H介于0.373 6~0.374 8)。进一步与宣和猪的肉质性状关联分析显示,G420A位点的GG基因型能显著提高肉色评分(P<0.05),GA基因型能显著提高熟肉率和降低失水率(P<0.05);A896G位点的AA基因型能显著提高熟肉率(P<0.05);A1114G位点和C1720T位点与滴水损失及肌内脂肪(IMF)含量存在显著关联(P<0.05)。此外,宣和猪的肉色评分、大理石纹和熟肉率均以ACAC单倍型组合的最高,失水率和滴水损失均以GTAC组合的最低,IMF含量以GTGT组合最高。不同单倍型组合与失水率、滴水损失和IMF含量存在显著或极显著关联(P<0.05或P<0.01)。研究结果初步揭示了宣和猪ENHO基因多态性与肉色评分、失水率、熟肉率、滴水损失和IMF含量显著关联,携带有AC单倍型的个体肉质水平相对较高。

摘  要:
To determine the association between ENHO gene polymorphism and meat quality traits in Xuanhe pigs, the SNPs of the ENHO gene sequences of 119 Xuanhe pigs were detected by direct sequencing of PCR products, and the genotype and haplotype combination of the SNP loci were correlated with the meat quality traits. The results showed that four SNPs variants were detected in the ENHO gene of the Xuanhe pigs, among which three SNPs(G420A, A896G and A1114G) were detected in the intron 1 region and one SNP(C1720T) was detected in the 3′-UTR region. Among the four SNP loci, A896G, A1114G and C1720T showed moderate polymorphism(PIC ranged from 0.497 1 to 0.499 7), and G420A showed low polymorphism(PIC=0.123 9) and low genetic diversity(H=0.123 9). The genetic diversity of the three loci was also relatively rich(H ranged from 0.373 6 to 0.374 8). Further correlation with the meat quality traits of Xuanhe pigs showed that the GG genotype at the G420A locus of the ENHO gene significantly increased the meat color score(P<0.05), the GA genotype significantly increased the cooked meat rate and decreased the water loss rate(P<0.05). The AA genotype at the A896G locus significantly raised the cooked meat percentage(P<0.05). The A1114G and C1720T sites were significantly correlated with the drip loss and IMF content(P<0.05). In addition, the meat color score, marbling score and cooked meat rate of the Xuanhe pigs were the highest in the ACAC haplotype combination, the water loss rate and drip loss were the lowest in the GTAC haplotype combination, and the IMF content was the highest in the GTGT haplotype combination. Different haplotype combinations had significant or extremely significant associations with water loss rate, drip loss and IMF content(P<0.05 or P<0.01). The above results indicated that the polymorphism of the ENHO gene had significant effects on the meat color score, water loss rate, cooked meat rate, drip loss and IMF content of Xuanhe pigs, and the meat quality traits of individuals carrying the AC haplotype were at a high level.
