裂殖壶菌营养成分及其多糖特征分析

摘  要:
为研究裂殖壶菌Schizochytrium sp.的脂质组成及多糖初级结构,采用气相色谱(GC)、发射光谱(ICP)、高效液相色谱(HPLC)和红外光谱(FTIR)等方法分析了裂殖壶菌基本组分和微量元素含量,研究了其脂质组成及多糖初级结构。结果表明:裂殖壶菌的水分、灰分、蛋白质、脂质、碳水化合物含量分别为1.49%、9.79%、15.49%、43.97%、29.26%;可溶性多糖提取率为2.13%,其多糖和硫酸根含量分别为49.81%和4.76%;裂殖壶菌富含镁、钙、钠金属元素;气相色谱分析脂肪酸组成发现,DHA是提取裂殖壶菌油脂中的主要脂肪酸,油脂中还含有EPA及其他饱和脂肪酸;高效液相色谱分析多糖的单糖主要以半乳糖为主,并含有少量的甘露糖、鼠李糖;红外光谱分析多糖的硫酸基团主要位于半乳糖残基的C6位。研究表明,裂殖壶菌中富含人体所必需的多不饱和脂肪酸DHA,具有较大的开发潜力。

摘  要:
The approximate components, mineral content, lipid profile and structural characteristics of polysaccharide were analyzed in a single-celled marine fungus Schizochytrium sp. by gas chromatography(GC), emission spectrum(ICP), high performance liquid chromatography(HPLC) and Fourier transform infrared spectroscopy(FTIR). The results showed that the fungus had 1.49% of moisture, 9.79% of ash, 15.49% of crude protein, 43.97% of lipid and 29.26% of carbohydrate. The soluble polysaccharide was extracted at a rate of 2.13%, with 49.81% of polysaccharide and 4.76% of sulfate. The analysis of mineral contents indicated that the fungus was rich in magnesium, calcium, sodium and other metal elements. The GC analysis revealed that docosahexaenoic acid(DHA) was the main fatty acid in the extracted lipids, and eicosapentaenoic acid(EPA) and other saturated fatty acids in the extracted lipids. HPLC analysis indicated that the monosaccharide was primarily comprised of galactose, and a small amount of mannose and rhamnose, and FTIR analysis showed that the sulfate group was mainly located in the C6 position of the galactose residue, indicating that Schizochytrium sp. has great development potential due to rich in polyunsaturated fatty acid DHA.
