紫草素对肝癌细胞增殖和凋亡的影响

摘  要：
为了研究紫草素对肝癌细胞增殖和凋亡的影响,试验以SMMC-7721为研究对象,利用倒置显微镜观察细胞的形态变化,通过体外细胞毒性试验(MTT)比色法和流式细胞仪检测紫草素处理前后对肝癌细胞株SMMC-7721增殖和诱导细胞凋亡的作用。结果表明:在紫草素作用下,SMMC-7721形态发生改变,细胞增殖受到抑制,细胞凋亡率为(19.00±1.31)%和(23.04±1.25)%。说明紫草素对肝癌细胞株SMMC-7721具有明显的凋亡诱导作用,为抗肿瘤药物的研究提供了理论基础。

摘  要：
In this research,SMMC-7721,a hepatoma carcinoma cell line was used to evaluate the inhibitory and apoptotic effects of shikonion on tumourigenesis,the morphological changes upon treatment were observed with phase contrast microscopy,cytotoxicity was analysed with MTT assay,and proliferative and apoptotic effects were detected with flow cytometry.The results suggested that upon treatment with shikonion the cells became irregular in morphology,were inhibited in growth and division,and exhibited apoptotic rates of(19.00±1.31)% and(23.04±1.25)%.This study proved that shikonion did have significant pro-apoptotic effects on SMMC-7721 line,and hence provided a theoretical basis for future development of prospective anti-tumor drugs.
