适合乳果糖生产的β-半乳糖苷酶高产株的筛选及培养基优化

摘  要:
通过紫外、超声和甲基磺酸乙酯(EMS)诱变乳酸克鲁维酵母(Kluyveromyces lactis),筛选出一株适合乳果糖生产的β-半乳糖苷酶高产菌株K1-16-7,其乳果糖产量达到44.88g/L。通过Plackett-Burman实验设计、中心组合实验设计对其摇瓶培养基进行优化,采用优化配方酶活可以达到147.6U/g。

摘  要:
The UV,ultrasonic and ethylmethane sulfonate(EMS)were used for mutating Kluyveromyces lactis.The high-β-galactosidase-yield strain K1-16-7 which was suitable for lactulose production was screened out.The yield of lactulose can reached 44.88g/L.The culture medium was optimizated by Plackett-Burman design and central composite design.After the optimization,the yield of β-galactosidase could reached 147.6U/g.
