摘  要:
家禽骨质特性是家禽重要的经济性状指标,研究显示肿瘤坏死因子α(TNF-α)对人类及动物骨代谢有显著影响。为了揭示影响家禽骨强度的主效基因,研究以体重较轻、体型较小的大围山微型鸡以及大型鸡种武定鸡为研究对象,和艾维因肉鸡作对比,通过检测骨强度以及骨骼中TNF-α基因的表达量,研究TNF-α基因的表达量对大围山微型鸡和武定鸡股骨强度的影响。结果显示,4~16周龄大围山微型鸡股骨强度均显著高于武定鸡和肉鸡(P<0.05),且武定鸡股骨强度高于肉鸡;每一阶段大围山微型鸡股骨软骨中TNF-α基因m RNA表达量显著低于其它两个鸡种(P<0.05);相关分析显示TNF-α基因的表达水平与股骨强度指标呈负相关。可见,TNF-α基因对家禽的骨强度有重要影响,是影响家禽骨强度的重要候选基因。

摘  要:
The bone property is an important economic indicators of poultry. It has been reported that tumor necrosis factor-α(TNF-α)is closely related to the growth and development of skeletal in animals and human. The objective of this study was to investigate the effect of TNF-α m RNA expression on the bone strength of Daweishan Mini chicken which is a small-size local chicken breed,and Wuding chicken which is a big-size local chicken breed and Avian broiler as control. The results showed that femoral bone strength of Daweishan Mini chickens from 4 to 16 weeks of age was higher than Wuding chicken and Avian broiler,and femoral bone strength of Wuding chicken was higher than Avian broiler. In contract,the expression of TNF-α m RNA in femoral cartilage from 4 to 16 weeks of age in Daweishan Mini chickens were significantly lower than the other two breeds(P<0.05). In addition,correlation analysis revealed that TNF-α m RNA expression in femoral cartilage was negative correlation with femoral bone strength. It was obvious that TNF-α gene played an important role in regulating bone growth and strength in chickens,and could be selected as a candidate gene of bone strength for molecular breeding in chicken.
